Summary Genes for the major storage protein of potato, patatin, have been mapped genetically and physically in both the potato and tomato genomes. In potato, all patatin genes detected by the cDNA clone pGM01 map to a single locus at the end of the long arm of chromosome 8. By means of pulsed field gel electrophoresis (PFGE) it was possible further to delimit this locus, containing 10–15 copies of the gene, to a maximum size of 1.4 million base pairs. Hybridizations with class-specific clones suggest that the locus is at least partially divided into domains containing the two major types of patatin genes, class I and II. In tomato, patatin-homologous sequences were found to reside at the orthologous locus at the end of chromosome 8. The approximately three copies in tomato were localized by PFGE to a single fragment of 300 kilobases. Whereas the class II-specific 5 promoter sequences reside in tomato at the same locus as the coding sequences, the single class I-specific copy of the 5 promoter sequences was localized on chromosome 3 with no coding sequence attached to it. A clone from this chromosome 3 locus of tomato was isolated and by restriction fragment length polymorphism mapping it could be further shown that a similar class I-specific sequence also exists on chromosome 3 of potato. As in tomato, this copy on chromosome 3 is not linked to a coding sequence for patatin. The results are discussed with respect to genome evolution and PFGE analysis of complex gene families.  相似文献

Abstract. The effect of growth temperatures on quantum yield (φ) was examined for leaves at different stages of development within the immature canopies of two crops of field grown maize ( Zea mays cv. LG11) sown on 3 May and 20 June 1990. During the period of 23 to 49d after sowing, the crop sown on the 3 May experienced temperatures below 10°C on 19 occasions compared with only two for the crop sown on 20 June. A period of severe chilling at the end of May and the beginning of June was associated with a marked reduction in φ for all leaves in the early-sown crop. This chill-induced depression in φ was greater in recently emerged than more mature leaves in the canopy and was found to be accompanied by modifications in the polypeptide profiles of thylakoids isolated from the leaves. During the chilling period, decreases in some polypeptides, notably in the range of 41–42 and 20kDa apparent molecular size, and increases of polypeptides of c. 15–16kDa were observed compared with leaves developing at warmer temperatures in July. The efficiency of converting intercepted radiation into dry matter (conversion efficiency) was 42% lower in the early- than late-sown crop, but no significant relationship between conversion efficiency and quantum yield was found in either treatment.  相似文献

Carbon isotope ratios (13C) for bubble CH4 in a submerged paddy soil were studied in Yokohama, Japan, throughout a growing period, and its variation was found. Bubble CH4 collected from other 33 paddy fields in Japan was also measured for its 13C and the results agreed with Yokohama. Furthermore, the variation occurred irrespective of the amount and the type of supplied organic substances to the fields (whole rice straw, rice stubble, or compost). The 13C value (average value of -55.9 ± 4.24) from these paddy fields was higher than those of the CH4 emitted from African and North American paddies. The higher value was little affected by their difference in the supplied organic substances. CH4 oxidation likely occurs for bubble CH4 in the shallow paddy fields. A rough estimate of the total CH4 production, using isotope mass balance, showed that 17 to 22% of organic carbon supplied to Japanese paddies transforms to CH4.  相似文献

Summary A single scan method for the suppression of signals arising from zero-quantum coherences (ZQC) is analysed with respect to its application to NMR experiments on proteins. The ZQC are dephased during a spinlock period due to the natural RF inhomogeneity of a commercial probe. A quantitative analysis of a ZQC-compensated NOESY experiment is given. Although the build-up curve for the cross peaks in ZQC-compensated NOESY experiments differ from those in uncompensated experiments, interproton distances in medium-sized proteins can be evaluated with high accuracy. The proposed method is compared with other techniques for ZQC suppression.  相似文献

Summary A rapid method for recording three-dimensional triple-resonance experiments utilising pulsed field gradient techniques is proposed, and applied to the HNCO experiment. In order to optimise the sensitivity of the method, a short phase cycle is used in conjunction with the pulsed field gradients to select the desired coherence transfer pathway. Summary Agapanthus umbelatus pollen tubes (PTs) display a number of different growth patterns when germinated in an electric field of 750 mV· mm–1. When pollen is germinated near the cathode (82.44% of orientation to the cathode side) or near the anode (55.35% of orientation to the anode), growth is oriented parallel to the applied field but when germinated at an intermediate position, there is random growth. An increase and decrease in the orientation rates as well as reversion of the polarized growth were observed when the growth conditions were systematically altered. These findings reflect the influence of different ionic currents present in the germination medium. These ionic currents induce the formation of ionic gradients, which were monitored by ion-HPLC. The individual omission of Ca2+, K+, Mg2+ and Cl suppresses or alters the oriented growth pattern. The presence of ionic gradients is not by itself suficient to trigger the polarization of tube growth as the presence of an electric field which drives the ionic currents is essential for this to occur.Abbreviations PT Pollen tube - DNS 3,5-dinitro salycilic acid; - TP transient polarization - HPLC high precision liquid chroma tography - DC direct current  相似文献

Abstract.
  • 1 The population dynamics of a natural population of Sympetrum danae (Sulzer) were studied, by means of a mark—release—recapture method, to find indications of dispersal by mature adults. Additional information on orientation and the influence of phenotype (including age), density and weather was obtained by observations under natural conditions and in a large outdoor field cage.
  • 2 80% of all mature adults caught around the natural pond were estimated to be immigrants. Considerable immigration was confirmed by the abdomen length of unmarked matures which did not accord with that expected from the locally emerged population. Total numbers captured and the minimum number known to be alive fell sharply at the time when most individuals were mature. At the same time, new matures continued to enter and disappear from the study area at an increasing rate.
  • 3 Dispersive behaviour (escape flights) in the field enclosure was primarily age dependent in both sexes: it occurred from an age of 25–30 days onwards. This coincides with the time-lag between the emergence peak and increased dispersal in the field. Secondary effects on escape flights suggested that males tended to abandon low density areas.
  • 4 Males flew relatively more southward than females.

Several species of weakly electric fish reflexively change their frequency of electric organ discharge (EOD) in response to sensing signals of similar frequency from conspecifics; that is, they exhibit jamming avoidance responses (JAR).Eigenmannia increases its EOD frequency if jammed by a signal of lower frequency and decreases its EOD frequency if jammed by a signal of higher frequency. This discrimination is based on an analysis of the patterns of amplitude modulations and phase differences resulting from signal interference. Fish of the closely related genus,Sternopygus, however, do not exhibit a JAR. Here we show that despite lacking this behavior,Sternopygus shares many sensory processing capacities withEigenmannia:
1.  Fish could be conditioned to discriminate the sign of the frequency difference (Df) between an exogenous sinusoidal signal and its own EODs with as few as 300 training trials.
2.  Fish can discriminate the sign of Df for jamming signals with an amplitude as low as 2 V/cm (p-p); which is approximately 40 dB below the amplitude of the fish's EOD, as measured lateral to the operculum.
3.  Fish appear to discriminate the sign of Df by evaluating modulations in signal amplitude and differences in the timing of signals received by different areas of the body surface.
4.  Sternopygus processes electrosensory information through band-pass modulation filters. Tuning to modulation rates over at least 4–16 Hz exists.

Waterstriders (Gerris paludum) often try to maintain a nearly stationary position on a moving water surface. Passive motion is restricted to 3 degrees of freedom: yaw, longitudinal, and transverse displacement. They correct for passive rotation and translation by distinct behavioral sequences. The compensatory behavior is predominantly visually controlled.
1.  When waterstriders are rotated and translated simultaneously, they are able to discriminate their own rotation and translation visually.
2.  They discriminate their rotation from their translation even if the visible pattern is restricted to a monocular visual field.
3.  They detect rotation only if they see an extended pattern.
The restriction of degreess of freedom reduces the complexity of the motion-induced visual flow field. Each motion component induces its own flow field component. We propose that those areas of the visual field are preferred for analysis where the directions of the 3 flow field components differ most. These areas (Figs. 2 and 3) have their largest extent at 45° above the horizon.  相似文献
